Abstract
Polymerase chain reaction(PCR) has had an opportunity for the rapid detection of
Mycobacterium leprae in clinical specimens for the diagnosis of leprosy. And the
development in immunoperoxidase staining has given the information of localization of
the bacilli or antigen in the pathological slides.
We performed PCR in 27 fresh-frozen, 16 paraffin-embedded tissues and 9 skin
smears from leprosy patients and also, immunoperoxidease staining(labelled streptavidin
biotin method) in 5 slides from multibacillary(MB) leprosy patients and 7 slides from
paucibacillary(PB) leprsoy patients, both of which did not demonstrate any bacilli.
As our results, the PCR methods seemed more sensitive in fresh-frozen tissues, skin
smear samples and parson-embedded tissues in order. BIs of used samples were than
3+. Under the peroxidase staining 3MB slides demonstrated specifically stained
cytoplasms of macrophages and 2 PB slides showed epithelioid cells and nerves and
some endothelial cells were specifically stained. We expect to use those methods for
detection of PB leprosy patients.
